Preparation, Purification and Performance Evaluation of Polyclonal Antibody Against SARS-CoV-2 Produced in Rat

Purpose: Among all known human coronaviruses, some viruses (e.g., SARS-CoV-2) cause severe pneumonia or even death. With the regard to its spread and importance of rapid identification/treatment, because pAbs are relatively cheap, able bind more sites on antigens neutralize them, this study was done for production purification anti-SARS-CoV-2 polyclonal antibodies (pAb) in rats. Methods: Viral antigen performed by PEG/NaCl precipitation. The efficiency sucrose cushion method also investigated produce a purer antigen. Immunization antibody ammonium sulfate precipitation (33%), dialysis, ion-exchange chromatography. Western blotting enzyme-linked immunosorbent assay (ELISA) were verify specificity. All data analyzed SPSS software. Results: results showed that amount concentrated virus increased with increase PEG concentration. Moreover, purity. Besides, induction immune response rats pAb high titers reached via these ELISA/western blot indicated suitable antibody-antigen interaction. Additionally, it shown chromatography could be technique IgG purification. Conclusion: Herein, we presented simple cheap whole viral particles quality. verified elicit good rat. obtained specificity toward SARS-CoV-2 antigens. Accordingly, proposes promising vaccine development.
